Microbiological strategies for the treatment of pulmonary infections caused by Mycobacterium avium complex and Mycobacterium abscessus

Objectives

  1. Implement a personalized antibiogram adapted to clinical isolates of Mycobacterium avium complex (MAC) and Mycobacterium abscessus (MAB) in patients with chronic lung disease and infection by these species
  2. Determine the minimum inhibitory concentration (MIC) and the minimum bactericidal concentration (MBC) of new/unusual antibiotics in M.avium (MAV) and MAB
  3. Determine the MIC/MCB of combinations of two new/unusual drugs and one reference in MAV and MAB
  4. Determine the MIC/CMB of the three-drugs combinations in MAV and MAB using the microdilution method and time-kill curves
  5. Analyse the biofilm production and determine the MIBC/MBEC of the three-drugs combinations in MAV and MAB 
  6. Characterize the clonal relationship in MAC and MAB isolates.

 

Methodology

MAC and MAB isolates from collection and MAC/MAB strains isolated from patients with chronic lung disease will be studied. The microdilution method will be used to design the personalised antibiogram and to study the MIC/MBC of antibiotics individually, and in two-drugs and three-drugs combinations. The three-drugs combinations will also be studied by the time-kill curves method. Biofilm production will be evaluated with the crystal violet method. The CMEB/CMIB will be determined after to treat the biofilm with the best three-drugs combinations from objective 4. Massive sequencing will be performed with the Illumina NextSeq system platform and the Fourier transform infrared spectrometry with the IR Biotyper equipment (Bruker).
