The general objectives of the proposal are to identify modifiable factors and brain mechanisms that act to increase:

  1. Resistance to Alzheimer’s disease (AD) pathologies - amyloid and tau - and/or
  2. Brain vulnerability/resilience - measured as loss/preservation of neural integrity - during midlife
  3. Assessing their association with cognitive change

The specific objectives of the present proposal are as follows:

1.1. To assess the associations of baseline and longitudinal changes on amyloid and tau with proximity to parental age at onset

1.2. To assess the role of modifiable factors on these associations

2.1. To develop an imagingbased approach to measure vulnerability/resilience

2.2. To quantify the effects of multiple modifiable factors on vulnerability/resilience measurements

3.1. To identify the combination of factors that best predict cognitive decline
