El microbioma intestinal y la disrupción circadiana

Un estudio epidemiológico molecular sobre enfermedades cardiometabólicas y salud mental

Objetivos

Escasos datos experimentales y en humanos muestran que la microbiota intestinal presenta ritmos circadianos y que los trastornos del sueño y las comidas a deshora pueden alterar su composición y función.

Evaluaremos la relación entre el desajuste circadiano y el microbioma intestinal.

 

Metodología

Realizaremos un estudio epidemiológico molecular en 200 trabajadores nocturnos y diurnos de dos hospitales de un estudio europeo del exposoma.

Incorporaremos al estudio el microbioma, con muestras fecales anuales, y un contacto tras 2 años. El ADN fecal se extraerá y secuenciará para determinar las abundancias relativas de las unidades taxonómicas operativas bacterianas. Examinaremos la diversidad y abundancia microbiana evaluando los índices de diversidad alfa y beta.

La salud cardiometabólica y mental se evaluarán al inicio y prospectivamente. Los biomarcadores incluyen la HbA1c, hormonas, biomarcadores de inflamación, longitud telomérica y marcadores epigenéticos. El sueño, la exposición lumínica y el horario de las comidas se evaluarán con sensores, aplicaciones y cuestionarios.

Se utilizarán modelos lineales mixtos y modelos de regresión logística de efectos mixtos para el análisis estadístico de los resultados continuos y binarios, respectivamente, ajustando por factores de confusión.

 

Resultados esperados

El estudio identificará las vías que asocian el desajuste circadiano con el microbioma intestinal y los efectos en la salud a largo plazo. Los resultados ayudarán a desarrollar programas de prevención necesarios en trabajadores nocturnos y en la población general.
